Il a \u00e9t\u00e9 rapport\u00e9 qu'elle inhibe l'activit\u00e9 de la tyrosinase par un m\u00e9canisme non comp\u00e9titif, r\u00e9duisant la conversion catalys\u00e9e par la tyrosinase de la L-tyrosine en L-DOPA et de la L-DOPA en dopaquinone \u2014 \u00e9tapes cl\u00e9s de la biosynth\u00e8se de la m\u00e9lanine. Elle peut \u00e9galement moduler les voies de m\u00e9lanogen\u00e8se li\u00e9es \u00e0 l'inflammation, y compris la signalisation associ\u00e9e \u00e0 la COX\/PGE\u2082 (Yokota et al., 1998).<\/p>\n\n      <p>Des recherches plus r\u00e9centes ont caract\u00e9ris\u00e9 davantage l'activit\u00e9 de signalisation en amont de la glabridine. Cela signifie que la glabridine agit \u00e0 plusieurs niveaux : en amont au niveau de la cascade de signalisation (r\u00e9duisant l'expression des enzymes d\u00e9pendantes du MITF) et au niveau enzymatique (inhibant directement l'activit\u00e9 catalytique de la tyrosinase).<\/p>\n\n      <p>Le r\u00e9sultat : moins de m\u00e9lanine est produite, ce qui entra\u00eene une r\u00e9duction de la teneur en m\u00e9lanine dans les m\u00e9lanosomes et une diminution de l'accumulation de pigment en aval.<\/p>\n\n      <h3>Niacinamide \u2014 Transfert des m\u00e9lanosomes<\/h3>\n\n      <p>La niacinamide (vitamine B3) agit en aval de la synth\u00e8se de m\u00e9lanine \u2014 \u00e0 l'\u00e9tape du transfert. Le m\u00e9canisme propos\u00e9 implique la modulation des voies de signalisation k\u00e9ratinocytes-m\u00e9lanocytes associ\u00e9es au transfert des m\u00e9lanosomes. En r\u00e9duisant ce transfert, la niacinamide limite la livraison des m\u00e9lanosomes aux k\u00e9ratinocytes, o\u00f9 ils contribuent \u00e0 la pigmentation visible (Hakozaki et al., 2002).<\/p>\n\n      <p>La niacinamide n'inhibe pas directement la tyrosinase ni la production de m\u00e9lanine.
